This course will give you practical insight into the most productive proteomics techniques: Manufacturing, staining, scanning and software assisted evaluation of high resolution 2D-gels as well as mass spectrometric protein identification of 2D-gel protein spots will be addressed. Hands-on experience can be gathered in crucial areas of 2D-gel handling, 2D-gel sample preparation and mass spectrometric protein identification.


Day 1

9:00 - 9:45
Seminar: Introduction into 2D-gel techniques

10:00 -12:30

Demonstration: Purging of 2nd dimension of large-gel 2D-gel technique, sample application onto 1st dimension tube gel (prepared samples), start of 1st dimension large-gel technique run.

13:30 -18:00

Demonstration: Small gel technique.
Demonstration: Purging of small gel technique 1st and 2nd dimension gels.Termination of 1st dimension large gel technique, application of 1st onto 2nd dimension. Start of 2nd dimension run.


Day 2

9:00 - 9:45
Seminar: Practical aspects of 2D-gel technique.

10:00 -12:30

Demonstration: Scanning of 2D-gel images.
Demonstration: Sample application onto 1st dimension small gel technique. Start of run.

13:30 -18:00

Supervised Training: Termination of 2nd dimension large gel technique and start of staining. Termination of 1st dimension small gel technique and application of tube gel onto 2nd dimension. Run, determination and start of staining of the 2nd dimension small gel technique.


Day 3

9:00 - 9:45
Seminar: Mass spectrometry for protein identification.

10:00 -12:30

Demonstration: Tryptic digest and sample preparation for mass spectrometry.

13:30 -18:00

Demonstration: Measurement of prepared standards.
Supervised Training: Sample preparation of 2D-gel plugs. Start of tryptic digest.


Day 4

9:00 - 9:45
Seminar: Analysis of mass spectrometry data and Mascot searching.

10:00 -12:30

Supervised Training: Sample preparation for mass spectrometry from 2D-gel plugs. Measurement of MS-spectra.

13:30 -17:00

Supervised Training: Measurement of MS-spectra (continued). Analysis of the MS-spectra.

17:00 -18:00

Discussion of results.
